Daewoo run comfortable buses in Pakistan, but if they don't run a service from Karachi to Larkana, I'm sure there are other bus companies which do. I was advised not to walk, and to take (shared) rickshaws from the station to the archeological site, which I did.An alternative way to get to Moenjodaro would be to use the nearest town, which is Larkana.

As I understand it, Moenjodaro is the more interesting site to visit - there's meant to be more to see there, and it has a reputation as the best preserved site of the Indus Valley Civilization (the only other similar site I have been to is Dholavira in India, which is also very interesting). I found Moenjodaro fascinating - it's an incredible place, an excavation of an incredibly complex city from millenia ago. Hi everybody, Makli Hill is about 3km before the town of Thatta itself, so if you're planning on visiting the cemetery there, you can get off the bus there rather than in Thatta itself.
The nearest village is Dokri, perhaps 4km or so from Moenjodaro.

You should find regular transport of some kind from Larkana to Dokri, and you can then take a rickshaw from Dokri to Moenjodaro.The only other place in Sindh I've been is Sukkur, which has a few interesting places to see and is worth a visit if you have the time. I can't imagine much in the transport situation has changed since then, but my information is a few years old so take it as you will. I caught the first bus of the morning, which I think left around 7am, but I can't remember the time for sure. You can contact me through that website, if you wish.
There are at least a couple of trains from Karachi which stop here. Karachi and Thatta are both in the same time zone Asia/Karachi. Make sure to visit the national museum while you're in Karachi - there's a better collection of artifacts from Moenjodaro in Karachi then there is at Moenjodaro itself.
